[gene] Cbl (named after Casitas B-lineage Lymphoma) is a mammalian gene encoding the protein CBL which is an E3 ubiquitin-protein ligase involved in cell signalling and protein ubiquitination. Mutations to this gene have been implicated in a number of human cancers, particularly acute myeloid leukaemia. ==Discovery== In 1989 a virally encod... Oncogene of a murine retrovirus that induces lymphomas and leukaemias. Protein has N-terminal transforming region with phosphotyrosine-binding (PTB) domain and C-terminal region with RING finger motif, large proline-rich region and a leucine zipper. Cbl serves as a substrate for receptor and non-receptor tyrosine kinases and binds to Grb2, Crk and p85 of PI-3-kinase. May act as a negative regulator of tyrosine kinase signalling. Signals to JNK through C3G. See also Table O1.
