Cloning strategy for PCR products that relies on the tendency of Taq polymerase> Taqpolymerase to add an extra dA at the 3&` end of newly synthesized DNA strands, thus leaving a single base 3&` overhang. Vectors are accordingly prepared with single base dT 3&` overhangs, allowing ligation of sticky ends.
